TNF Alpha Gene and Chronic Inflammation | Decoding Inflammation

The Cytokine That Mobilizes Everything
TNF-α does not merely initiate inflammation. It coordinates it. Released by macrophages, monocytes, and T cells in response to infection, injury, or oxidative stress, TNF-α activates NF-κB, triggers apoptosis in damaged cells, recruits neutrophils to sites of injury, and signals downstream cytokine cascades to amplify the inflammatory response.
In acute disease, this is essential biology. The body that cannot mount a robust TNF-α response cannot contain infection or resolve injury. The body whose TNF-α signaling persists beyond the acute phase, however, inhabits a state of chronic low-grade systemic inflammation that accelerates tissue aging across every organ it bathes.
The -308G/A Variant
The most extensively studied TNF-α polymorphism is the -308G/A substitution, rs1800629, located in the promoter region of the TNFA gene. The A allele creates a binding site that increases transcription factor affinity, resulting in constitutively elevated TNF-α production. AA homozygotes produce significantly more TNF-α at baseline than GG carriers. GA heterozygotes occupy the functional middle ground.
A comprehensive 2025 meta-analysis in Frontiers in Immunology confirmed that -308A allele carriers show elevated serum TNF-α across rheumatoid arthritis, type 2 diabetes, nonalcoholic fatty liver disease, and major depressive disorder cohorts. The variant is not disease-specific. It is an inflammatory amplifier that operates across every system the immune response touches.
The Interaction With NF-κB Variants
TNF-α and NF-κB form a positive feedback loop: TNF-α activates NF-κB, and NF-κB drives further TNF-α transcription. In carriers of both TNF-α promoter variants and NF-κB pathway polymorphisms, this loop is both more easily activated and more difficult to extinguish. The Oxygen Free Radical Crisis operates precisely within this architecture, oxidative stress triggers TNF-α, which amplifies inflammation, which generates more oxidative stress.
The GENEFLAMMATION protocol treats TNF-α status as a central inflammatory regulatory variable. Therapeutic strategies for high-TNF genomic profiles target the NF-κB axis directly: curcumin bioavailability optimization, omega-3 dosing calibrated to FADS1 genotype, and resolution pathway support through specialized pro-resolving mediators.
