MTHFR Gene and Chronic Inflammation | Decoding Inflammation

What MTHFR Actually Does
Methylenetetrahydrofolate reductase, MTHFR, encodes the enzyme that converts folate into its active form, 5-methyltetrahydrofolate. This molecule donates methyl groups across hundreds of biochemical reactions. When the enzyme underperforms, the methylation cycle slows. Homocysteine accumulates. The body shifts toward a pro-inflammatory state.
The two most studied variants are C677T and A1298C. The C677T variant, when homozygous, reduces enzyme activity by approximately 70 percent. That is not a minor inefficiency. It is a structural bottleneck at the heart of cellular metabolism.
Why This Matters for Inflammation
Elevated homocysteine activates NF-kB, the master inflammatory transcription factor. It promotes oxidative stress by depleting glutathione, the body's primary antioxidant. It impairs endothelial function, stiffening blood vessels and accelerating cardiovascular risk.
The Oxygen Free Radical Crisis, the framework Dr. Rivera uses to explain the upstream origin of all chronic inflammatory disease, begins here. MTHFR dysfunction is not the only driver, but in carriers of the TT genotype, it is often the dominant one.
What the Research Shows
A 2024 meta-analysis in Nutrients confirmed that C677T homozygosity is associated with significantly elevated plasma homocysteine even in subjects with adequate dietary folate intake. Supplementing with standard folic acid does not correct the problem because the enzyme cannot convert it. The active form, methylfolate, bypasses the block entirely.
Pharmacogenomic data from PharmGKB further shows that MTHFR status modifies response to methotrexate, antiepileptic drugs, and several antidepressants. This is precision medicine made concrete.
What This Means for Your Genome
If you carry the C677T or A1298C variant, your methylation machinery is running at a deficit. That deficit touches neurotransmitter synthesis, DNA repair, detoxification, and immune regulation simultaneously. The GENEFLAMMATION protocol reads your MTHFR status alongside a panel of related SNPs, MTRR, CBS, BHMT, DNMT3A, to build a methylation map specific to your genome. From that map, a therapeutic roadmap follows.
